Vykon\u00e1vaj\u00ed funkci bun\u011b\u010dn\u00e9 elektr\u00e1rny, jeliko\u017e v\u00a0nich prob\u00edhaj\u00ed d\u016fle\u017eit\u00e9 biochemick\u00e9 pochody.<\/strong><\/p>\n<p>Nejd\u016fle\u017eit\u011bj\u0161\u00edm z\u00a0nich je vznik adenosintrifosf\u00e1tu (ATP) pomoc\u00ed bun\u011b\u010dn\u00e9ho d\u00fdch\u00e1n\u00ed. Molekuly ATP slou\u017e\u00ed jako zdroj energie pro dal\u0161\u00ed bun\u011b\u010dn\u00e9 procesy. Krom\u011b toho se mitochondrie pod\u00edlej\u00ed na produkci tepla, jsou sou\u010d\u00e1st\u00ed metabolismu tuk\u016f i l\u00e1tek steroidn\u00ed povahy a hraj\u00ed v\u00fdznamnou roli p\u0159i bun\u011b\u010dn\u00e9m r\u016fstu \u010di programovan\u00e9 bun\u011b\u010dn\u00e9 smrti (apopt\u00f3ze).<\/p>\n<p>Mitochondrie maj\u00ed sv\u016fj vlastn\u00ed genom. D\u00edky tomu se mitochondrie ozna\u010duj\u00ed jako tzv. semiautonomn\u00ed organely. Ka\u017ed\u00e1 eukaryotick\u00e1 bu\u0148ka tak obsahuje jadernou i mitochondri\u00e1ln\u00ed DNA (mtDNA), a\u010dkoliv mtDNA p\u0159edstavuje asi jen 0,1 % ve\u0161ker\u00e9 bun\u011b\u010dn\u00e9 genetick\u00e9 informace.<\/p>\n<p>Mitochondri\u00e1ln\u00ed genom je velice n\u00e1chyln\u00fd na vznik mutac\u00ed. Proto se v\u00a0lidsk\u00e9 populaci vyskytuje n\u011bkolik variant mtDNA, kter\u00e9 se rozd\u011bluj\u00ed do haploskupin a dal\u0161\u00edch podskupin. Mitochondri\u00e1ln\u00ed haploskupiny jsou vyu\u017e\u00edv\u00e1ny p\u0159i evolu\u010dn\u00edch a genealogick\u00fdch anal\u00fdz\u00e1ch. mtDNA se toti\u017e d\u011bd\u00ed v\u00fdhradn\u011b po mate\u0159sk\u00e9 linii (tedy z\u00a0matky na jej\u00ed potomky) a d\u00edky tomu \u00a0je mo\u017en\u00e9 podle haploskupiny ur\u010dit geografick\u00fd p\u016fvod p\u0159edk\u016f po mate\u0159sk\u00e9 linii.<\/p>\n<p>P\u016fvodn\u00ed mitochondri\u00e1ln\u00ed haploskupina je haploskupina L (tzv. mitochondri\u00e1ln\u00ed Eva) vyskytuj\u00edc\u00ed se na \u00fazem\u00ed Afriky. V\u00a0Evrop\u011b maj\u00ed nej\u010dast\u011bj\u0161\u00ed v\u00fdskyt haploskupiny H, J, K, T, V, X a U. V\u00a0Asii se nej\u010dast\u011bji vyskytuj\u00ed haploskupiny F, C, W, M, D, N, K, U, T, A, B, C, Z\u00a0a U a v\u00a0Austr\u00e1lii haploskupiny M, Q, S, O, N, nebo P.<\/p>\n<p>Mutace mtDNA vedou k\u00a0v\u00e1\u017en\u00e9mu naru\u0161en\u00ed energetick\u00e9 bun\u011b\u010dn\u00e9 produkce i k tk\u00e1\u0148ov\u00e9 dysfunkci. D\u00edky mutac\u00edm v\u00a0mitochondri\u00e1ln\u00edm genomu bylo identifikov\u00e1no n\u011bkolik mitochondri\u00e1ln\u00edch onemocn\u011bn\u00ed s\u00a0nejr\u016fzn\u011bj\u0161\u00edmi projevy. Mezi onemocn\u011bn\u00ed asociovan\u00e9 s\u00a0mutacemi v\u00a0mitochondri\u00e1ln\u00edm genomu pat\u0159\u00ed nap\u0159. mitochondri\u00e1ln\u00ed myopatie, Leberova heredit\u00e1rn\u00ed optick\u00e1 neuropatie (LHON), Leigh\u016fv syndrom, matern\u00e1ln\u011b d\u011bdi\u010dn\u00fd diabetes a hluchota atd.<\/p>\n<p>Znalost vlastn\u00ed mitochondri\u00e1ln\u00ed haploskupiny m\u00e1 praktick\u00fd v\u00fdznam, jeliko\u017e n\u011bkter\u00e9 haploskupiny jsou spojovan\u00e9 s\u00a0vy\u0161\u0161\u00edm rizikem v\u00fdskytu n\u011bkter\u00fdch onemocn\u011bn\u00ed (t\u0159eba pr\u00e1v\u011b LHON nebo Parkinsonovy \u010di Alzheimerovy choroby), ale i s\u00a0adaptac\u00ed na \u017eivotn\u00ed prost\u0159ed\u00ed.<\/p>\n<p>Mutace v\u00a0mitochondri\u00e1ln\u00edm genomu a metabolismus mitochondri\u00ed jsou bun\u011b\u010dn\u00e9 procesy \u010dasto spojovan\u00e9 se st\u00e1rnut\u00edm organismu, i kdy\u017e celkov\u00fd proces st\u00e1rnut\u00ed je zat\u00edm jen m\u00e1lo prozkouman\u00fd. P\u0159irozen\u00e9 st\u00e1rnut\u00ed je typick\u00e9 poklesem v\u00a0bun\u011b\u010dn\u00e9m metabolismu a sn\u00ed\u017een\u00e9 regenera\u010dn\u00ed schopnosti tk\u00e1n\u00ed. Tyto zm\u011bny doprov\u00e1z\u00ed i pokles mitochondri\u00e1ln\u00ed kvality a aktivity. Spolu s mutacemi mtDNA m\u016f\u017ee doch\u00e1zet i k\u00a0rozvoji mnoha r\u016fzn\u00fdch onemocn\u011bn\u00ed spojen\u00fdch s\u00a0p\u0159ib\u00fdvaj\u00edc\u00edm v\u011bkem.
