Na po\u010d\u00e1tku \u017eivota je jedin\u00e1 bu\u0148ka, kter\u00e1 se vz\u00e1p\u011bt\u00ed za\u010d\u00edn\u00e1 nep\u0159etr\u017eit\u011b d\u011blit a specializovat, aby dala vzniknout specifick\u00fdm tk\u00e1n\u00edm a org\u00e1n\u016fm. Jednotliv\u00e9 bu\u0148ky v\u00a0lidsk\u00e9m t\u011ble se vz\u00e1jemn\u011b li\u0161\u00ed; maj\u00ed r\u016fzn\u00e9 fyziologick\u00e9 funkce, fyzik\u00e1ln\u00ed vlastnosti a slo\u017een\u00ed sv\u00fdch stavebn\u00edch kamen\u016f \u2013 organel.<\/p>\n<p>Bu\u0148ky spolu interaguj\u00ed v\u00a0z\u00e1vislosti na sv\u00e9 funkci a prost\u0159ed\u00ed, ve kter\u00e9m se vyskytuj\u00ed. M\u00edsto bu\u0148ky si m\u016f\u017eeme p\u0159edstavit dosp\u011bl\u00e9ho \u010dlov\u011bka, kter\u00fd se jinak chov\u00e1 v\u00a0pr\u00e1ci k pod\u0159\u00edzen\u00fdm a jinak doma ke sv\u00fdm bl\u00edzk\u00fdm. \u00a0Modulac\u00ed sv\u00e9ho chov\u00e1n\u00ed a jedn\u00e1n\u00ed tak m\u016f\u017ee do jist\u00e9 m\u00edry ovliv\u0148ovat my\u0161len\u00ed a postoje lid\u00ed kolem sebe. \u00a0Stejn\u011b tak funguj\u00ed i bu\u0148ky. P\u0159\u00edkladem jsou rakovinn\u00e9 bu\u0148ky, kter\u00e9 reguluj\u00ed funkci a chov\u00e1n\u00ed bun\u011bk v\u00a0okol\u00ed. Proto je nanejv\u00fd\u0161 d\u016fle\u017eit\u00e9 v\u010das identifikovat tyto \u201e\u0161patn\u011b se chovaj\u00edc\u00ed\u201c malign\u00ed bu\u0148ky, aby bylo mo\u017en\u00e9 zabr\u00e1nit jejich v\u00fdvoji v metastatick\u00e9 rakovinn\u00e9 bu\u0148ky.<\/p>\n<p>Docent Powell \u00a0(Garvan Institut of Medical Research,\u00a0Darlinghurst, Sydney, Australia) vysv\u011btluje, \u017ee klasifikace bun\u011bk se obvykle prov\u00e1d\u00ed identifikac\u00ed povrchov\u00fdch bun\u011b\u010dn\u00fdch marker\u016f pomoc\u00ed technologie zvan\u00e9 pr\u016ftokov\u00e1 cytometrie. Nedostatkem t\u00e9to metody je fakt, \u017ee navzdory identick\u00fdm povrchov\u00fdm znak\u016fm mohou bu\u0148ky reagovat rozd\u00edln\u011b. Profesor Powell uv\u00e1d\u00ed jako d\u016fkaz pro sv\u00e9 tvrzen\u00ed zm\u00edn\u011bn\u00e9 rakovinn\u00e9 bu\u0148ky, kter\u00e9 na pr\u016ftokov\u00e9m cytometru vypadaj\u00ed stejn\u011b, ale pouze jejich \u010d\u00e1st m\u00e1 potenci\u00e1l metastazovat.<\/p>\n<p>Pro pochopen\u00ed chov\u00e1n\u00ed bun\u011bk mus\u00edme j\u00edt hloub\u011bji. Ka\u017ed\u00e1 bu\u0148ka organismu obsahuje stejn\u00fd soubor gen\u016f, ale ne v\u0161echny jsou aktivn\u00ed. Pr\u00e1v\u011b zap\u00edn\u00e1n\u00edm a vyp\u00edn\u00e1n\u00edm jednotliv\u00fdch gen\u016f a regulac\u00ed jejich aktivity doch\u00e1z\u00ed k\u00a0diferenciaci bun\u011bk a rozd\u00edln\u00fdm reakc\u00edm na podn\u011bty z okol\u00ed.<\/p>\n<p>T\u00fdm docenta Powella vyvinul novou techniku pro identifikaci jednotliv\u00fdch typ\u016f bun\u011bk, tzv. \u201escPred\u201c. \u201escPred\u201c je zalo\u017eena na sekvenov\u00e1n\u00ed jednotliv\u00fdch molekul RNA a umo\u017e\u0148uje sn\u00edmat aktivitu gen\u016f pouze v jedn\u00e9\u00a0dan\u00e9 bu\u0148ce. Anal\u00fdzou z\u00edskan\u00fdch dat bylo detekov\u00e1no obrovsk\u00e9 mno\u017estv\u00ed informac\u00ed o\u00a0rozd\u00edlech mezi jednotliv\u00fdmi bu\u0148kami. Na z\u00e1klad\u011b t\u011bchto informac\u00ed byla zah\u00e1jena tvorba \u201eknihovny lidsk\u00fdch bun\u011bk\u201c, aby byla mo\u017en\u00e1 snaz\u0161\u00ed identifikace stejn\u00e9ho typu bun\u011bk u r\u016fzn\u00fdch lid\u00ed.<\/p>\n<p>Ve sv\u00e9m p\u0159\u00edsp\u011bvku publikovan\u00e9m v\u00a0\u010dasopise Genome Biology v\u011bdci p\u0159edlo\u017eili nad\u011bjn\u00e9 v\u00fdsledky sv\u00e9 studie. P\u0159i anal\u00fdze vzork\u016f bun\u011bk kolorekt\u00e1ln\u00edho karcinomu se jim poda\u0159ilo identifikovat rakovinn\u00e9 bu\u0148ky s p\u0159esnost\u00ed v\u00edce ne\u017e 98%. Metoda \u201escPred\u201c neumo\u017e\u0148uje pouze d\u0159\u00edv\u011bj\u0161\u00ed detekci onemocn\u011bn\u00ed. Informace o\u00a0aktivit\u011b jednotliv\u00fdch gen\u016f mohou pomoci i s\u00a0v\u00fdb\u011brem vhodn\u00fdch l\u00e9k\u016f a l\u00e9\u010debn\u00fdch postup\u016f.
